Tylostoma was named by Sharpe (1849) [Sepkoski's age data: J Kimm T Dani Sepkoski's reference number: 914]. Its type is Tylostoma torrubiae.

It was assigned to Gastropoda by Acquaotta (1929); to Naticidae by Kase (1984); to Neotaenioglossa by Sepkoski (2002); and to Tylostomatidae by Squires and Saul (2004) and Kollmann (2009).
