Glypharthrus was named by Raasch (1939) [Sepkoski's age data: Cm Fran Cm Trep-u Sepkoski's reference number: 1031]. Its type is Eurypterus thomasi. It was considered monophyletic by Lerosey-Aubril et al. (2017).
It was assigned to Merostomata by Sepkoski (2002); to Aglaspidida by van Roy (2006); and to Aglaspididae by Størmer (1955), Hesselbo (1992), Ortega-Hernandez et al. (2013) and Lerosey-Aubril et al. (2017).
