Amphitryon was named by Hawle and Corda (1874). Its type is Amphitryon murchisonii.

It was corrected as Amphytrion by Harrington et al. (1959).

It was assigned to Remopleuridinae by Harrington et al. (1959) and Nikolaisen (1983); to Asaphida by Sepkoski (2002); and to Remopleurididae by Owen (1981) and Jell and Adrain (2002).
