Polytryphocycloides was named by Smith and Paul (1982) [Sepkoski's age data: D Fras]. Its type is Polytryphocycloides huronensis. It was considered monophyletic by Smith and Paul (1982).

It was assigned to Cyclocystoididae by Smith and Paul (1982); and to Isorophida by Sepkoski (2002).
