Mesophthirus was named by Gao et al. (2019). Its type is Mesophthirus engeli. It is the type genus of Mesophthiridae.
It was assigned to Coccoidea by Grimaldi and Vea (2021); to Mesophthirinae by Shcherbakov (2022); and to Mesophthiridae by Gao et al. (2019) and Gao et al. (2022).
