Desmatotherium was named by Scott (1883).

It was synonymized subjectively with Helaletes by Osborn (1889), Hay (1902), Troxell (1922), Radinsky (1963) and Schoch (1984).

It was assigned to Lophiodontidae by Scott (1883); to Hyrachyinae by Peterson (1919); to Helaletinae by Osborn (1923); to Ceratomorpha by Prothero and Schoch (1989); to Ceratomorpha by Scott (1883), Matthew (1909), Schoch (1989) and Colbert and Schoch (1998); and to Helaletidae by Hough (1955) and Bai et al. (2017).
