Pentamplexus was named by Schindewolf (1940) [Sepkoski's age data: P Leon P Dora Sepkoski's reference number: 893]. It is not extant.

It was assigned to Tachylasmatinae by Hill (1956); to Pentaphyllinae by Hill (1981); to Rugosa by Sepkoski (2002); and to Polycoeliidae by Fedorowski (2009).
