Karydomys was named by Theocharopoulos (2000). Its type is Karydomys symeonidisi.

It was assigned to Democricetodontini by Mörs and Kalthoff (2004); to Cricetinae by Maridet et al. (2011); and to Democricetodontinae by Theocharopoulos (2000) and López-Antoñanzas et al. (2019).

C. D. Theocharopoulos 2000Medium to large cricetids. Cusps inflated. Small posterosinus in the M1-M2. Ectlophs well developed in the M1-M2 of Karydomys symeonidisi and Karydomys boskosi, but absent in Karydomys zapfei. M3 and m3 very much reduced. Anteroconid low and blade-like and placed very close to the protoconid and metaconid.
