Protostrea was named by Chen (1976) [Sepkoski's age data: Tr Anis Sepkoski's reference number: 1066]. Its type is Ostrea sinensis.

It was assigned to Ostreidae by Chen (1976) and Vokes (1980); to Pterioida by Sepkoski (2002); and to Dimyidae by Fang et al. (2009) and Ros-Franch et al. (2014).
