Desmatosuchus was named by Case (1920). Its type is Desmatosuchus spurensis. It is the type genus of Desmatosuchidae.

It was assigned to Desmatosuchia by Case (1922); to Desmatosuchidae by Case (1920), Huene (1922) and Hay (1930); to Aetosauridae by Charig (1967); to Stagonolepididae by Parrish and Carpenter (1986), Carroll (1988), Small (1989), Parrish (1993), Long and Murry (1995), Heckert and Lucas (1999) and Irmis (2005); to Desmatosuchinae by Huene (1942), Heckert and Lucas (2000), Zeigler et al. (2002), Parker (2007) and Parker (2008); and to Desmatosuchini by Parker (2016).
