Mesturus was named by Wagner (1862) [Sepkoski's age data: J Toar J Kimm Sepkoski's reference number: 452,1066]. Its type is Mesturus verrucosus.

It was assigned to Pycnodontidae by Woodward (1895), Gardiner (1967); to Gyrodontidae by Wenz (1967); to Mesturidae by Nursall (1996), Nursall (1999), Poyato-Ariza and Wenz (2002); and to Pycnodontiformes by Sepkoski (2002).
