Kuphus was named by Guettard (1770). It is extant. Its type is Serpula polythalamia.

It was assigned to Annelida by Collignon and Cottreau (1927); and to Kuphinae by Vokes (1972), Woodring (1982), Ward (1992), Bouchet et al. (2010).
