Acaenoplax was named by Sutton et al. (2001). Its type is Acaenoplax hayae. It was considered monophyletic by Sutton et al. (2001).

It was assigned to Mollusca by Sutton et al. (2001); to Aplacophora by Sutton et al. (2012); and to Heloplacidae by Sutton et al. (2025).
