Amphitryon was named by Hawle and Corda (1874). Its type is Amphitryon murchisonii.

It was corrected as Amphytrion by Harrington et al. (1959).

It was assigned to Remopleuridinae by Harrington et al. (1959), Nikolaisen (1983); to Remopleurididae by Owen (1981), Jell and Adrain (2002); and to Asaphida by Sepkoski (2002).
